JSK Therapeutics is a clinical-stage oncology company developing first-in-class nitric oxide (NO)-generating anti-cancer therapeutics with improved safety profiles. Its lead compound JS-K is an arylated diazeniumdiolate prodrug that releases nitric oxide upon interaction with glutathione in a reaction catalyzed by glutathione-S-transferase — an enzyme expressed at significantly higher concentrations in cancer cells compared to normal tissue — enabling targeted delivery with fewer systemic side effects. JS-K was developed through collaboration between Dr. Larry Keefer at the National Cancer Institute and Dr. Paul Shami's laboratory at the University of Utah. The company received NIH SBIR funding to conduct pre-clinical toxicology studies in support of an IND application to the FDA.
